Molecular profiling of resistance alleles in Pectinophora gossypiella (Saunders) (Lepidoptera: Gelechiidae) collected from different locations

Abstract Background After the commercialization of insect-resistant transgenic Bt cotton Bollgard I & II, India ranks first in world production. Cotton insecticide consumption was drastically reduced as nearly 95% area replaced with II. However, benefits appear to have been diminished pink bollworm, Pectinophora gossypiella (Saunders) (Lepidoptera: Gelechiidae) developed field resistance Cry1Ac expressing 2008. In 2015, there an unusual survival P. on II some parts Gujarat and Maharashtra, which is a cause for concern. Results present study, PCR analysis gel visualization BGII resistant population showed that presence r1, r2, r3, r1s, r2s, r3s, ss mutated cadherin alleles, produced amplicon sizes 750 bp, 700 730 12,700 980 600 bp 1600 respectively, when seven different types allele(s) specific primers were used. The r1 mutant allele detected Amaravati, Wardha, Yavatmal, Nagpur, but not Akola Buldhana, using r1disfor Int-540. r2 Akola, r2disback Cad2366. r3 all locations r3disback Cad3221. Amplicons obtained respectively. amplicons 1270 r3s.The absence bands alleles individuals indicated possibility having allele, confirmed Cad3324 Int-651. observed field-collected populations from BG various locations, size same primers. Conclusions revealed Vidarbha provide evidence field-evolved cotton.
